I think part of why this is creepy is that the eyes are so different from each other, even if you ignore the colours. The red eye is much clearer and sharp, while the green is somewhat cloudy and dull. It's like a poorly matched glass eye, and that's definitely unsettling.

Just you wait till you meet an actual person who has complete heterochromia. It's even more unsettling to see it in real life...
