I understand the joke but there is any deeper conection? Do these two goddess even share a pantheon in any of their sincretisms?
They don't share a pantheon, but Wikipedia would suggest they at least probably wouldn't get along, as while they do overlap in some areas (associations with justice for example), they're opposites in other areas. Astraea is apparently a virgin goddess of purity (and innocence), while Ishtar is a goddess of love (and war) who had many lovers (she's noted as not being associated with marriage).
I don't know that much about Fate anymore, but I can still say with 99.8% confidence it has to do with whosebodies those goddesses are occupying.
Oh, I don't doubt for a moment that is part of it. I just think that the raito is a little less absolute given that when it comes to Ishtarin, Ishtar is running that show more so than Rin is. That said it is quite the possible that Ishtar "hates" Astraea as much as Rin and Luviagelita are known to fight and bicker with each other.
Given their FGO classes, I'd say Luviastraea would probably win here.
Just like with their first encounter.
Ah, my lovely forklift lady have finally made it into the game. Shame about her art though, I was really expecting her to LOOK like Luvia. I’m disappointed in myself that I couldn’t recognize her at first glance and I was really not expecting her to be a 4star.

She's a 4* yes, but she's also permanently added to the summon pool opposed to Rin's 5* limited form, which means you can only get Ishtar from banners and events.

That said she definitely doesn't have the skills and stats of a 4*. She's pretty goddamn jacked with good skills, passives and stats that makes her a fantastic choice for bosses, including fighting with any of the Rins. I'm fairly convinced she's only a 4* because of the collaboration she's released in, so Reines pretty much demanded being the 5*.
Also regardless of the art normally, her FA is nice to look at, so there's that.

The Goddesses were modeled in their Type-moon incarnations after Rin Tohsaka and Luvia Edelfelt from Fate/Stay Night, who are vicious rivals who have come to physical blows in a wrestling match. That's who these are.
