It would certainly help so long as they didn't turn Naruko into a girly girl and have a Naruko/Sasuke romance as one of the major plotlines.
Seriously though, I'd love to have a shonen series where the lead is actually a girl with "typical shonen lead" character traits. It'd be a nice change of pace from all these recycled main characters we've had in the shonen genre for ages.

If a girl like that was the lead in a shounen then it wouldn't be a shounen...it would be a shoujo with shounen elements. Naruto is a shounen with shoujo elements..
The whole point of shounen manga is that it's aimed for boys, with boys at the center. Boys generally like 'those types of character traits' just like some girls like being the 'lonely somewhat attractive girl surrounded by cute boys with hidden past' trait generally seen in shoujo plots.
However over the years alot of these type of shounen manga have been adopting alot of shoujo themes (y'know, deep meaning plots, more emotional drama etc). If Naruko was female and judging by Naruto's existing personalty then she would be a hyper active girly girl that can switch to a badass ninja. In short, she'd be Sakura, badass being severely optional.

IMO yes, given Naruto's personality when he's normal and then his sexy no jitsu personality, then there's Sasuke and how Naruto acts around him, add estrogen.
It may not be completely genderswap with Sakura but it's pretty damn close.

Its Easy for a Male character to have that lovable idiot stubborness But it would had been great to see a female take that character type the way the give it to Naruto
and even his liking for Sakura, playing on lesbian love angle with out the creepiness,(See Bleach) just a genuine "She is smart and pretty" because I think Sakura was at the start of the series, just annoying how its was "hearts and love" with her to Sasuke
And Narutos HATE for Sasuke, what better way to make a Girl even weirder but then by Hating the most popular boy in the whole school, and not in the tsundere way, Still the "I Must be better Then Him!" approach, and yet Still have that respect for him Naruto has because he see him more as a brother figure then a friend/rival
Would had made that scene where he was pushed/tipped into kissing Sasuke even more justifiable and funnier when Naruto played off the "How Gross!" moment afterwards
Even Hinata secretly admiring Naruto for that "never give up" attitude still would had made a nice angel if seeing that in another female, Love or no love there
The only issue there is the heaviness of GirlonGirl Love which people may had an issue with
Tried the novels and a bit of the NEXT anime. Lina's a good character to base a female protagonist on but she isn't quite what I was going for.
